Using Frozen Sperm

Sperm arrive in 0.5 ml thin walled tubes containing 250μl of sperm suspension on dry ice. Store sperm in a -80°C freezer.

To use the sperm, squeeze the eggs from three females as separate groups into a single large petri dish. Add room temperature molecular biology grade water (250μl) to the sperm suspension and fertilise immediately even though the sperm suspension may not be completely thawed. The fertilisation rate decreases fast so speed is important. Mix the eggs and sperm then allow 5 minutes prior to flooding with 0.1X MBS.
