H3K9ac-Mintbody is a modification-specific intracellular antibody (mintbody) composed of an scFv against H3K9ac fused to enhanced Green Fluorescent Protein (eGFP). Under control of the CMV promoter, the mintbody is ubiquitously expressed. Upon acetylation of H3K9, the H3K9ac-Mintbody specifically binds to the modified histone and accumulates in the nucleus, resulting in increased nuclear eGFP fluorescence. Thus, the dynamics of histone acetylation and active chromatin status can be monitored in vivo by quantifying nuclear eGFP intensity.
